主要研究成果:
1. 鱼类克隆研究:1970年代在童第周教授指导下,完成了鲤鱼和鲫鱼间的细胞核移植;1990年代领导完成了以去核金鱼卵为受体克隆转基因鲤鱼、以去核斑马鱼卵为受体克隆稀有鮈鲫的研究,实现了低等脊椎动物异种间克隆。
2. 转基因鱼研究:1984年首创鱼类基因转移研究。构建了“全鱼”基因转移表达载体,相继培育了快速生长转基因鲫鱼、泥鳅和鲤鱼,揭示了外源基因整合的动力学过程,并深入研究了转基因鱼的生态效应及生物安全问题,为鱼类基因育种奠定了理论基础。 此外,克隆了鱼类GH基因,研究其在脊椎动物早期演化的特殊意义。 发表研究论文100多篇,6次获国家和省部级科技成果奖励。
目前研究方向: 1. 主持北京大学遗传与发育生物学研究中心,以斑马鱼、果蝇、线虫等模式动物为材料,探索动物发育调控的分子机制以及相关的疾病模型的研究:规模性筛选和鉴定模式动物发育相关基因;研究基因表达图式及其在发育中的功能和可能的致病机制;从分子水平研究细胞分化与发育的调控机制。.
2. 以水生脊椎动物为材料,通过显微操作技术,探索发育过程中的核质关系以及胞质因子对发育和形态建成的影响。
Research Achievements As a co-principal investigator, we succeeded in nuclear transplantation between different genera in fishes in 1970’s. We pioneered a new research area of fish genetic engineering by producing the first batch of 2.3-3.4-fold-faster growing transgenic fishes in 1985. Based on the discovery of the instability and mosaic integration of foreign gene in the host genome, we established a model of generation of transgenic fish and propounded a strategy of producing homogeneous transgenics. By cloning of 4 genes from Cyprinidae, we were the first to propound and achieve an "all-fish" recombinant gene construct. We have been working comprehensively on the biosafety. Research work in my group provides theoretical and practical basis for application of genetic engineering to fish directional breeding. He also discovered the DNA marker for Cyprinus carpio and pointed out the significant structure of fish GH genes to the study of the early development of vertebrates.
Research Field
1.In the Center of Genetics and Developmental Biology at Peking University, zebrafish, fruit fly and worm are used to study the molecular mechanism behind embryogenesis and to be models of bridging gene regulation disorder and disease occurrences.
2.The technique of micromanipulation is employed to study the developmental interaction between nucleus and cytoplasm and the effects of cytoplasmic factors on the development and pattern formation in aquatic vertebrates.
